Detailed study of heterogeneous subpopulations of myeloma cells and their precursors/progenitors in monoclonal gammopathy patients can lead to identification and characterisation of myeloma stem cells. Using polychromatic flow cytometry with sorting of cells, phenotype of myeloma stem cells can be detected and different subpopulation can be selected for clonality assessment. In vitro culturing then can prove self renewal capacity of these myeloma stem cells and their multidrug resistance as well. We suppose that using these approaches we are able to obtain new information in area of pathophysiology of myeloma cells and their behaviour, which can be potentially used for determination of predictive factors applicable in progression of monoclonal gammopaty of undetermined significance to multiple myeloma.
