Medium 21: HAEMOPHILUS MEDIUM

    PPLO-Agar (Difco B412)        34 g 
    Horse serum                   50 ml 
    Glucose                        1 g 
    Yeast extract*               100 ml 
    Distilled water              900 ml 
 
Suspend PPLO-Agar and glucose in cold distilled 
water and heat to boiling. Sterilize at 121°C for 
15 min. Cool to 50°C, add aseptically the horse 
serum and yeast extract, mix well and pour into 
sterile Petri dishes. 
 
    *Yeast extract:
    Baker's yeast                250 g
    Distilled water             1000 ml 

Mix thoroughly and heat to boiling. Filter through
 paper filter, adjust pH to 8.0 and filter through
 Seitz filter. Maintain at -20°C. Before use check
 the sterility!
